背景:弥漫性B细胞淋巴瘤(DLBCL)是成人患者中最常见的非霍奇金淋巴瘤亚型,全球年发病率为25.0%至40.0%。尽管如此,该疾病的预后仍然很差。目的:迫切需要新的、可靠的用于预后预测的生物标志物。方法:利用基因表达Omnibus数据集中的449个DLBCL样本,检测了与嗜铁相关的长链非编码rna (lncRNAs)之间的关系。在应用单变量Cox分析排除与预后相关的lncRNAs之前,我们使用Pearson相关分析筛选了大量与铁死亡相关的lncRNAs。结果:为了预测DLBCL的预后,对11个与铁死亡相关的lncRNAs进行了选择算子Cox回归和最小绝对收缩。此外,研究表明,6种与铁衰相关的lncRNAs在建立预测风险模型方面最有效。DLBCL患者根据他们的中位风险评分被分为高风险组和低风险组。分层分析表明,采用11种与铁死亡相关的lncRNAs构建的模型具有更高的预后评估能力。在高危患者中观察到肿瘤相关通路的显著富集。年龄、世界卫生组织分级以及与铁死亡相关的lncRNA预后因素在创建nomogram时被考虑在内。结论:总而言之,生成的nomogram可以准确预测两个队列中DLBCL患者的总生存期。
