目的:探讨跨膜蛋白16A(TMEM16A/ANO1)在乳腺癌中的表达及其对癌细胞增殖、周期、凋亡、侵袭及体内成瘤的影响。方法:收集20例乳腺癌及配对癌旁组织检测TMEM16A表达;MCF-7细胞中敲低TMEM16A后检测增殖、周期、凋亡与侵袭;裸鼠移植瘤模型观察体内成瘤并检测MAPK通路蛋白及Cyclin D1。结果:乳腺癌组织中TMEM16A mRNA和蛋白均高表达(P<0.01)。敲低TMEM16A可抑制MCF-7细胞增殖、诱导G0/G1期阻滞、削弱侵袭(P<0.01),对凋亡影响不明显;移植瘤生长明显减缓(P<0.05),伴p-p38、p-ERK1/2及Cyclin D1下调,总蛋白无明显变化。结论:TMEM16A在乳腺癌中高表达,敲低后可能经p38/ERK1/2-Cyclin D1轴抑制肿瘤细胞增殖、侵袭及体内成瘤,是潜在治疗靶点。
