研究表明,源自牙周膜(PDL)细胞的高迁移率族蛋白 B1(HMGB1)可能是正畸牙齿移动和牙周组织重塑过程中的潜在调节因子。本研究旨在采用Waldo 法对小鼠磨牙进行持续加力,探究小鼠正畸牙齿移动过程中牙周组织中HMGB1的表达情况。实验选用6只7周龄的C57BL6 小鼠,将弹性带置于右上颌第一和第二磨牙之间的牙缝中,施加机械力3天后,用含4%多聚甲醛的0.1M磷酸缓冲液(pH7.4)经心脏灌注固定小鼠,然后取出上颌骨进行组织化学分析。组织学检查显示,张力侧出现局部牙周膜撕裂,压迫侧形成广泛的无细胞透明带。HMGB1的免疫定位在牙周膜张力侧、根尖区和牙髓中显著呈现,而在牙周膜压迫侧,由于透明带的存在,HMGB1的标记几乎检测不到。综上所述,我们得出结论,采用Waldo法进行的正畸牙齿移动会导致与螺旋弹簧法不同的组织学变化和HMGB1 表达模式,包括牙周膜撕裂和广泛的透明带形成,这可能会严重破坏牙周组织,进而阻碍牙齿移动。
