背景:长链非编码RNA(Long non-coding RNA,LncRNA)在肝细胞癌(hepatocellular carcinoma,HCC)的发生发展中发挥重要作用。本研究旨在探讨HOTAIR在HCC中的表达模式及其调控肿瘤转移的具体机制。方法:采用Northern blot检测临床样本中HOTAIR的水平;通过Western blot分析HOTAIR、DNMTs与PTEN之间的调控关系;利用划痕试验和Transwell实验评估细胞的迁移与侵袭能力。结果:HOTAIR在肝癌组织中显著高表达。HOTAIR能够上调DNMT1和DNMT3A的表达,进而抑制抑癌基因PTEN的水平。过表达HOTAIR显著增强了肝细胞的迁移和侵袭能力,而这一效应可被DNMT1的敲低所逆转。回复实验证实,敲低DNMT1可显著逆转HOTAIR引起的PTEN下调及细胞迁移能力的增强。结论:HOTAIR通过DNMT1/PTEN轴促进肝癌的进展,可能作为肝癌治疗的潜在靶点。
