雪莲注射液调控巨噬细胞极化改善心肌缺血再灌注损伤的实验研究

目的:探讨雪莲注射液调控巨噬细胞极化改善心肌缺血再灌注损伤的作用。方法:50只健康雄性C57BL/6小鼠随机分为假手术组、模型组、雪莲注射液低、中、高剂量组。假手术组开胸、穿线但不结扎冠状动脉,模型组和雪莲注射液低、中、高剂量组通过结扎冠状动脉构建MIRI模型。雪莲注射液低、中、高剂量组注射相应浓度的雪莲注射液,假手术组和模型组注射等体积0.9%氯化钠注射液。连续干预7d后,流式细胞术(FCM)测定M1、M2型巨噬细胞占比,酶联免疫吸附(ELISA)法检测血清TNF-α、IL-6、IL-10、TGF-β水平。结果:与假手术组相比,模型组小鼠的M1型巨噬细胞比例及M1/M2比值显著升高(P<0.01),而M2型巨噬细胞比例显著降低(P<0.01);同时,血清促炎因子IL-6、TNF-α水平显著升高(P<0.01),抗炎因子IL-10、TGF-β水平显著降低(P<0.01)。与模型组相比,雪莲注射液各剂量组可显著逆转上述变化,表现为M1型巨噬细胞比例及M1/M2比值降低(P<0.01),M2型巨噬细胞比例升高(P<0.01);同时,IL-6与TNF-α水平显著下降(P<0.01),而IL-10与TGF-β水平显著升高(P<0.01)。结论:心肌缺血再灌注损伤小鼠可促进巨噬细胞向M1型极化,并增加炎症反应,雪莲注射液可使巨噬细胞向M2型极化,进而缓解炎症反应。
