DCAF(DDB1-CUL4-associated factor)蛋白家族作为CRL4泛素连接酶复合物的关键底物识别亚基,通过介导靶蛋白的特异性泛素化修饰,广泛参与细胞周期调控、DNA损伤修复、信号转导及基因表达等关键生物学过程。近年研究表明,多种DCAF成员在肿瘤中呈现异常表达,并通过失调的泛素化降解途径驱动肿瘤发生、进展及治疗抵抗。本综述系统梳理了DCAF家族核心成员(如DCAF1、DCAF2、DCAF7、DCAF8、DCAF12、DCAF13等)在肝癌、结直肠癌、胃癌、多发性骨髓瘤等恶性肿瘤中的表达特征、分子机制及临床意义,探讨其作为新型诊断标志物和治疗靶点的潜力,并展望该领域面临的挑战与未来研究方向。
