目的:本研究旨在探讨miR-24-3p通过调控MTHFR对人结肠癌细胞凋亡作用研究。方法:生物信息学预测与miR-24-3p有结合的靶基因并采用双荧光素酶报告基因实验验证miR-24-3p与靶基因之间的靶向关系。用RT-qPCR分别检测miR-24-3p和靶基因MTHFR在结肠癌组织和细胞中的表达丰度。用凋亡试剂盒和蛋白印迹分别检测细胞凋亡情况和凋亡蛋白表达水平。结果:miR-243p在结肠癌组织和细胞中表达下调,而MTHFR则表达上调,且二者存在负调控关系。miR-24-3p与MTHFR能够结合并负向调控后者的mRNA和蛋白表达水平。过表达miR-24-3p或干扰MTHFR都能够促进结肠癌细胞凋亡,同时过表达二者结肠癌凋亡比例下降。结论:miR-24-3p通过下调MTHFR的表达,促进结肠癌细胞凋亡。
